History:
(Savage Tales I#1 (fb) - BTS / others (see comments)) - Wanting the "Super-Soldier" serum being designed by Ted Sallis, the subversive organization AIM (Advanced Idea Mechanics) conspired with a bitter Ellen Brandt, whom Ted had neglected since their honeymoon, to obtain it.
(Savage Tales I#1 (fb) - BTS) - After Sallis completed his serum, he made arrangements for Hamilton to pick it up.
(Savage Tales I#1 (fb) - BTS) - Alan and his associate presumably slew Hamilton and waited at his cabin for Sallis.
(Savage Tales I#1 - BTS) - When Hamilton failed to show up for their meeting, Sallis -- unwilling to take any chances and knowing he had one dose of completed/distilled serum, as well as the formula committed to his memory -- burnt his written records on the formula.
(Savage Tales I#1) - Sallis opened the door to Hamilton's cabin and saw Hamilton lying dead on the floor. Alan and his associate then came out of hiding and confronted Sallis at gunpoint, after which Ellen revealed her true loyalties. Refusing to surrender the serum, Sallis punched Alan, but his associate then restrained Sallis in a double arm bar, allowing Alan to punch Sallis in the abdomen repeatedly. As his associate noted that Sallis had had it, Sallis instead threw his head back, smashing the associate in the chin. Breaking free, Sallis then grabbed Alan's head and pulled it down as he smashed his knee into Alan's chin or neck, after which Sallis got into his car and fled in his car.

prevent enemy agents from obtaining the serum, Sallis injected the
serum into himself, after which he crashed his car through a fence and
sank into the swamp.
(Savage Tales I#1) - As Sallis' car sank, Ellen cursed Alan and his associate for having allowed them to lose everything, Alan suggested they could get a winch truck out in the morning. Ellen noted that the sign reported the lake (swamp) to be bottomless, and they headed back in their car.
(Savage Tales I#1 - BTS) - Sallis was mutated by both the serum and the Nexus' energies into the monstrous Man-Thing.
(Savage Tales I#1) - Despite
its distorted perceptions and memory, the
Man-Thing traveled back to the road and intercepted their car. Alan's
associate slammed on the brakes to avoid a collision, after which the
Man-Thing grabbed Alan and burnt his face and/or strangled him
(regardless leaving him incapacitated on the ground), after which it
picked Alan's associate over its head and then broke his back of its
knee. The Man-Thing further burned and scarred Ellen's face before
wandering off.

I don't have all references for all of the ret-con information that's been added to the Man-Thing's origin. There was no mention of SHIELD, AIM, Project Gladiator (or Calvin or Morse or anyone else), Project Sulfur, the Super-Soldier serum, or anything else in the original story, but if Sallis was involved with all of those things, Hamilton may been as well. I'm starting a lengthy (probably at least a few years) project of profiling the Man-Thing (Ted Sallis), and I'll try to sort out all of the original references to all of those things as I go.
